RESULTS OF CHRONIC TOXICITY STUDY OF PROTEIN-SALT HYPEROSMOLAR SOLUTION

Abstract
Aim. The objective of this study is to investigate the effect of the new protein-salt infusion solution AXAM on the body weight dynamics and some hematological and biochemical parameters of the blood of white rats upon repeated administration, as part of the study of its chronic toxicity.
Materials and methods. The infusion solution under the laboratory code AXAM was investigated. The drug contains donor albumin (5%), pentatomic alcohol xylitol (5%), an alkalizing component – sodium acetate (0.26%), antihypoxant malate (0.067%), as well as electrolytes Na+ , K+, Ca++ and Cl–. Due to its composition, the total theoretical osmolarity of the drug is approximately 642 mOsm/l, pH 6.2-7.4. The study was conducted on male albino rats. The experimental group of animals was intraperitoneally injected with the drug AXAM at a dose of 10 ml/kg daily for 30 days. The intact animals were included in the control group. After the study, blood samples were taken from the animals for hematological and biochemical tests.
Results: It was established that after the administration of the drug AXAM, the weight gain of the experimental group of animals was statistically significant and amounted to +21.4% (p<0.05). Multiple infusions of the drug did not significantly affect the concentration of hemoglobin, color index, erythrocyte and leukocyte counts, as well as the leukocyte formula. Compared to the data of the control group, practically all biochemical parameters of the experimental group did not differ statistically significantly (p>0.05). Only the decrease in the concentration of calcium and uric acid was statistically significant (p<0.05). The glucose concentration was statistically significantly higher in the experimental group than in the control group (6.86±0.30 vs. 4.55±0.33 mmol/l, respectively).
Conclusions. Prolonged (for 30 days) intraperitoneal administration of the protein-salt infusion solution AXAM at a dose of 10 ml/kg body weight did not cause any toxic effects on the general condition, behavior, and weight gain of the animals, nor did it cause statistically significant changes in the hematological parameters and most of the biochemical parameters of the blood (with the exception of the levels of calcium and uric acid). Repeated administration of AXAM had an effect on the blood glucose level of the experimental animals (the glucose concentration in the experimental group was statistically significantly higher than in the control group).
